达氟沙星(Danofloxacin)在鸡体内的组织残留 被引量:6

摘要 建立了鸡组织中达氟沙星 ( Danofloxacin)的高效液相色谱 ( HPL C)测定方法 ,并测定了鸡单次内服 (每千克体重5 m g)给药后组织中的达氟沙星残留特征。组织样品用含 0 .0 15 mol/ L H3PO4 和 0 .0 15 mol/ L HCl O4 的水 -甲醇 ( 5 0∶5 0 )提取 ,5 0℃水浴酸性水解 90 min,离心后取上清液作 HPL C检测。色谱柱为 Hypersil BDS C1 8柱 ;荧光检测器检测 ,激发波长 2 80 nm,发射波长 4 4 0 nm;流动相为含 0 .0 15 m ol/ L 四丁基溴化铵的水 -乙腈 ( 90 5∶ 95 ,组织检测时为 915∶85 )混合物 ,85 %磷酸调 p H为 3.0。血浆、肝脏、肾脏、肺脏、肌肉中达氟沙星的检测限为 0 .0 2 m g/ L或 0 .0 2μg/ g,血浆样品回收率大于 90 % ,组织样品回收率均大于 74 %。鸡单次内服甲磺酸达氟沙星后 ,在 6 0 h内各组织中均可检出药物 ,但在 4 8h所有组织中药物残留量均低于相应的 MRL s(最高残留限量 )。结果表明 。 A HPLC method was established for determination of danofloxacin residues in chicken tissues.The drug in tissues was extracted with a methanol perchloric phosphoric solution,and the drug in plasma extracted with methanol.After centrifugation,the supernatant was used for analysis.Danofloxacin was separated on a Hypersil BDS C 18 column.The mobile phase consisted of acetonitrile and 0 001 5 mol/L tetrabutylammonium bromide(85/915) was adjusted pH3 0 with phosphoric acid.A fluorescence detector was utilized with an excitation wavelength of 280 nm and an emission wavelength of 440 nm.The recovery of danofloxacin in 4 tissues(liver,kidney,lung,muscle) was greater than 74%,the detection limit was 0 02 μg/g in tissues and 0 02 mg/L in plasma.The residue depletion of danofloxacin in 4 tissues of chicken was investigated following single oral administration(5 mg/kg).After administration 48 h,the danofloxacin could be detected in 4 tissues,but the levels were below their correlative maximum residues limits(MRLs=0 4 μg/g or 0 2 μg/g).The results indicated the elimination of danlfloxacin from tissues was relatively slow.
