摘要
番鸭细小病毒莆田株 ( MPV-P)和鹅细小病毒莆田株 ( GPV-P)分别感染的番鸭胚尿囊液经氯仿处理、PEG沉淀和 Sepharose 4 B柱纯化。纯化样品在电镜下观察 ,均见到实心和空心 2种病毒粒子。病毒粒子呈圆形 ,立体对称 ,无囊膜 ,直径为 2 0~ 2 4 nm。经 SDS-PAGE分析 ,MPV和 GPV均呈现 3条结构蛋白带。 MPV结构蛋白的相对分子质量约为 VP1890 0 0、VP2 780 0 0和 VP3 6 1 0 0 0 ,其中 VP3 为主要结构蛋白。GPV的相对分子质量与 MPV有微小差别。 MPV和 GPV核酸均为单链 DNA,长度约为 5 1 0 0 0 bp。分别以 MPV核酸和GPV核酸为模板 ,加到无引物的 DNA聚合酶 大片段合成体系中 ,合成产物经 1 %琼脂糖凝胶电泳 ,均可见长度约为 5 6 0 0 bp的双链 DNA带 ,证明 MPV和 GPV核酸的 3′末端具有发夹结构。对这 2种病毒核酸及经 Klenow合成的双链核酸进行限制性内切酶分析 ,两者的酶切结果明显不同 ,表明 MPV和 GPV核酸在一级结构上存在明显差异。以上结果证明 ,MPV和 GPV不但在致病性上有显著差异 。
The allantic fluids of duck embryo inoculated with muscovy duck parvovirus(putian strain) and goose parvovirus(putian stain) were extracted with chloroform,pricipitated with PEG and followed by sepharose 4B column chromatography.The purified MPV and GPV Contained full and empty non enveloped cosahedral viral particles with a diameter of 22 24 nm in size.SDS PAGE analysis revealed that MPV have 3 major structural polypeptides,they are VP 1 89 000 ,VP 2 78 000 and VP 3 61 000 ,and that of GPV are very little different from MPV.The nucleic acids of Both MPV and GPV are single stranded DNA with about 5 100 bp in size.The restriction endonuclease digestion showed the very diffenent pattern between MPV and GPV nucleic acids.The results indicated that significant differences exist between the genome primary structure of MPV and GPV.
