从以腹泻和呼吸困难为主要症状的雏番鸭肝、脾中分离到2株番鸭细小病毒(Muscovy Duckling Parvovirus,MPV)MPV-F和MPV-P。番鸭胚胎接种病毒后8~7天死亡,胚体呈弥漫性充血、出血。病毒能在番鸭胚肾(MDEK)细胞和番鸭胚成纤维(MDEF)细胞上增殖。雏番鸭人工感染病毒后出现与自然罹病者相似的症状,并回收到病毒。小鹅和雏半番鸭接种病毒后不出现临床症状。病毒耐乙醚、胰蛋白酶、酸和热。但是,对紫外线照射敏感。在电镜下病毒呈晶格排列,有实心和空心两种病毒粒子,直径为20~24nm,无囊膜。有4种结构蛋白;VP1(85kD)、VP2(70kD),VP3(56kD)和VP4(39kD),其中VP3是主要结构蛋白。病毒核酸为单链DNA,分子量约为5.1kb。酶联免疫吸附试验(ELISA),荧光抗体试验(FA)、胶乳凝集(LPA)试验、琼脂扩散沉淀(AGP)试验和中和试验(NT)中,MPV的单抗与鹅细小病毒(Gosling Parvovirus,GPV)不发生交叉反应,GPV单抗也不与MPV发生反应。证明MPV是雏番鸭以泻、喘为主要症状的疫病的病原,是细小病毒科的一个新成员。
Two strains of Muscovy Duckling Parvovirus ( MPV-F and MPV-P ) were isolated from the liver and spleen of Muscovy Ducklings (MD ) with symptoms of diarrhea and gasping on Muscovy Duck farms in Fuji-an during 1988. Muscovy Duck embryo inoculated with MPV died in 3-7 days with massive congestion and hemorrhage. Muscovy ducks inoculated with MPV showed pathology similar to that of field cases, and MPV was recovered from the tissues of these ducks. But goslings and semi-Muscovy ducks inoculated with MPV did not show any symptom. MPV could be propagated in Muscovy Duck embryokidney and fibroblast cell cultures. It resisted acid ( pH 3.0), heat ( 60℃ 60min. ), either, chloro-form and trypsin, but was sensitive to ultraviolet rays. By electron mi-croscopy, the virions consisted of spherical full and empty particles 20-24nm in diameter, with no envelope and arranged in partially crystal-line distribution. SDS-PAGE revealed that the virus contained 4 struc-ture polypeptides, VP1 ( 85kD ) ,VP2 ( 70kD ) ,VP3 ( 56kD ) and VP4(39kD), with VP3 as the major structural protein. The virai nucleic acid of MPV was sensitive to S1 Nuclease & DNase, but insensitive to RNase. Elec trophoresis of virai DNA in nondenatured agarose gels with the A-DNA Hindi digest markers showed that size of virai DNA is about 5.1kb. In tests of ELISA, FA, LPA, AGP and NT, monoclonal antibodies ( Mc Ab ) against MPV reacted only with MPV,a McAb against Goslings Par-vovirus (GPV ) reacted only with GPV. The results indicates that MPV is the pathogen of the infectious disease of Muscovy ducks characteri-zed mainly by diarrhea and gasping and is a new member of Parvoviridae.
