重组人乳铁蛋白对口腔癌细胞周期及周期因子表达影响的实验研究 被引量:8

Effects of Recombinate Human Lactoferrin on Cell Cycle and Expression of CyclinD1,p19 and p27 in Oral Cancer Cells

摘要 目的:探讨重组人乳铁蛋白(Recombinate human lactoferrin,rhLF)对口腔癌Tca8113细胞周期及周期调控因子cyclin D1、p19和p27基因表达的影响。方法:分别采用流式细胞仪和RT-PCR方法分析细胞周期及周期调控因子cyclin D1,p19和p27的mRNA水平。结果:50μg/mL、100μg/mLrhLF作用Tca8113细胞后,细胞增殖减慢,G0/G1期细胞明显增多,与对照组相比差异具有统计学意义(P<0.05);Tca8113细胞的cyclin D1 mRNA表达下降显著,50μg/mL和25μg/mLrhLF实验组与对照组相比差异具有统计学意义(P<0.05);p19和p27m RNA表达水平的变化不明显。结论:rhLF通过下调口腔癌细胞周期蛋白cyclinD1的表达来调控周期阻滞,为乳铁蛋白用于肿瘤的预防及治疗提供实验基础。 Objective: To study the effects of lactoferrin on cell cycle and expression of cyclinD1, p19 and p27 in Tca8113 cells. Methods: Flow cytometry (FCM) was used to determine the effect of rhLF on cell cycle. The gene expression of cyclinD1, p19 and p27 were examined by RT--PCR. Results: The percent ages of G0/G1 phase cells of 50 8g/rnl, 100 μg/ml rhLF groups were significantly higher than that of the control groups(P〈0.05), mRNA expressions of CyclinD1 in 50 μg/ml and 25 μg/ml rhLF experimental groups are significantly different compared with the control group (P〈0.05). The level of p19, p27 mRNA expression did not change obviously. There was no significant difference compared with the control group. Conclusion: Lactoferrin modulates directly cell cycle ar- rest by down--regulation of cyclin D1.
