摘要
目的检测黏液表皮样癌组织中β-连环素(catenin)和细胞周期蛋白(cyclin)D1的表达。方法采用免疫组化SP法检测30例黏液表皮样癌组织中β-catenin、cyclinD1的表达。结果β-catenin在正常涎腺组织中主要分布于涎腺导管上皮、腺泡上皮和肌上皮细胞。cyclinD1在正常涎腺组织中无表达。黏液表皮样癌组织β-catenin10例(33.3%)呈正常膜表达,20例(66.7%)β-catenin胞质和(或)胞核内异位表达;黏液表皮样癌组织中16例(53.3%)呈cyclinD1过度表达。β-catenin呈正常膜表达的10例中,2例(20.0%)呈现cyclinD1过度表达;而β-catenin呈异常表达的20例中,14例(70.0%)cyclinD1过度表达。黏液表皮样癌β-catenin的异常表达与cyclinD1的过度表达呈正相关性(P<0.05)。结论β-catenin的异常表达可能通过促使或激活cyclinD1的过度表达导致黏液表皮样癌的发生和发展。
Objective To examine the expression of β-catenin and cyclinD1 in mucoepidermoid carcinoma to evaluate their function in the formation and progression. Methods The alterations of β-catenin and cyclin D1 in 30 cases of mucoepidermoid carcinoma and 10 sections of normal salivary gland were determined using highly sensitive SP immunohistoehemi method. Results β-catenin expressed on the duct, acinus and epithelial cell in 10 sections of normal salivary gland. CyclinD1 was not detected on the normal salivary gland. It was only observed in 10/30(33.3% ) of these cases with normal membranous staining of β-catenin. 20 cases(66.7% ) showed abnormal staining of β-catenin. 16 sections(53.3%) showed over-expression of cyclin D1. There was a significantly positive correlation between the abnormal expression of catenin and over-expression of cyclinD1 ( X^2 = 0. 480, rs = 0.321, P 〈 0.05). Conclusion Down regulation of β-catenin accumulation in the cytoplasm/nuclear may promote malignant transformation and progression by triggering cyclinD1 over-expression in mucoepidermoid carcinoma.
