Expression and Significances of TGFβ_1,Smad4 and CyclinD_1 in Human Oral Squamous Cell Carcinoma.

摘要 目的:探讨TGF-β1、smad4及cyclinD1在口腔鳞癌发生发展过程中的作用。方法:采用免疫组化SP法分析TGF-β1、smad4、cyclinD1在正常口腔黏膜和口腔鳞癌组织中的表达情况。结果:TGF-β1蛋白阳性表达率在正常口腔黏膜组织为35.7%,口腔鳞癌组织中为69.0%(P<0.05);smad4在口腔鳞癌组织中的表达率为54.7%,明显低于口腔正常黏膜85.7%(P<0.05);TGF-β1、smad4表达的改变与口腔鳞癌淋巴结转移有关;smad4和cy-clinD1的表达呈负相关(P<0.05)。结论:TGF-β1的过表达与口腔鳞癌侵袭转移紧密相关;smad4的失表达不仅有利于提高细胞对TGF-β1的生长抵抗,也增强了口腔鳞癌的侵袭性;cyclinD1过表达与口腔鳞癌的发生和发展密切相关。。 Objective: To investigate the role of TGF -β1, smad4 and cyclinD1 expression in oral squamous cell carcinoma and normal oral mucosa. Methods: SP immunostaining technique was used to examine the expression of TGF -β1, smad4 and cyclinD1 in oral squamous cell carcinoma and normal oral mucosa. Results: TGF-β1 expressed positively in 35.7% of normal oral mucosa while 69.0 % of oral squamous cell carcinoma ( P 〈 0.05 ). Smad4 expressed positively in 54.7% of oral squamous cell carcinoma, which was lower than that of normal oral mucosa ( P 〈 0.05 ). The expression of TGF -β1 and smad4 in oral squamous cell carcinoma was associated with carcinoma lymph node metastasis. Negative correlation was observed between the expression of smad4 and that of cyclinD1 ( P 〈 0.05 ). Conclusion: The overexpression of TGF -β1 was highly related to the invasion or metastasis of oral squamous cell carcinoma. Decreased expression of smad4 was helpful not only to enhance cell growth resistance to TGF -β1 but increase the invasion of oral squamous cell carcinoma. The overexpression of cyclinD1 was closely correlated with occurrence and development of oral squamous cell carcinoma.
