流式细胞术分析牛乳铁蛋白素对Jurkat细胞增殖、分化的影响 被引量:1

Effects on the proliferation and differentiation of Bovine Lactoferrcin to Jurkat cell by flow cytometric analyses

摘要 采用流式细胞术分析了牛乳铁蛋白素(Bovine Lactoferrcin,LfcinB)对Jurkat细胞增殖周期、细胞表面分化抗原表达水平的影响。结果表明:LfcinB处理Jurkat细胞后,随着LfcinB浓度增加,处于G1/G0期的Jurkat细胞百分率明显上升,S期的细胞百分率明显下降,细胞表面分化抗原CD11b表达水平明显升高;此外,利用CCK-8法、NBT还原反应观察LfcinB对Jurkat细胞增殖、分化的影响,发现细胞增殖抑制率显著上升,NBT还原反应OD值明显提高,具有剂量和时间依赖性。以上结果说明,LfcinB能够通过细胞周期阻滞抑制Jurkat细胞增殖,并能诱导其分化。 In the experiments, flow cytometric analyses were used to research the effects of LfcinB on Jurkat cell cycle and cell surface differentiation antigen expression level. The results showed that after LfcinB treating Jurkat cell, as LfcinB concentration increasing, the cell percentage in G1/G0phase was increased significantly, and the cell percentage in S phase was decreased significantly, as well as cell surface differentiation antigen CD11b expression level improved obviously;in addition, the effects of LfcinB on cell proliferation and differentiation were observed by CCK-8 method and NBT reduction reaction, and found that cell proliferation inhibition rate increased significantly, and the OD value of NBT reduction reaction improved obviously, in a dose-dependent and time-dependent manner.The results above indicated that LfcinB could block cell cycle in G1/G0phase to inhibit Jurkat cell proliferation and could induce Jurkat cell differentiation.
