摘要
苜蓿根瘤菌AS1.159经亚硝基胍和Co ̄(60)γ射线诱变,获得一株高积累聚β-羟基丁酸(PH8)突变株J5177,积累的PHB可达细胞干重的30%,约为原出发菌株的10倍。采用分批补料发酵法,积累的PHB可达细胞干重的71%。菌体生长最适C/N比值为2.5,菌体积累PHB最适C/N比值为15;CaSO_4,Na_2MoO_4对菌体积累PHB有促进作用。
A mutant strain J 5177 with a high yield of poly-β-hydroxybutyric acid from Rhizobium melitoti AS 1.159 was obtained by chemical mutation using N-methyl-N'-nitro-soguanidine(MNNG)and treatment with Co_(60) radiation. The content of PHB obtained in batch culture was 30%,which was about 10 times that of the originel strain’s,The optimal combination of carbon source and nitrogen source for the growth of the strain was 2.5,and PHB accumulation was 15.There was the promotion of CaSO_4 and Na2MoO_4 to PHB accumulation in batch culture. The optimal medium volume was 125 mL in a 500 mL flask,The maximum content of PHB was 71%,which was obtained by fed-batch fer-mentation providing supplementation of glucose.
