摘要
为了解低分子甲壳胺多糖的抗肿瘤辅助疗效 ,作者利用高效液相色谱法 (HPLC)结合薄层色谱法 (TLC) ,探讨了SUGAR KS 80 2寡糖分析柱分析甲壳胺寡糖的条件 ,并尝试了几种薄层层析展开剂对甲壳胺寡糖组分的展层条件。结果表明 ,在HPLC流动相为 0 .0 0 1mol/LNaOH溶液 ,流速 0 .2mL/min时 ,可将混合的标准甲壳胺寡糖成分很好地分离 ;采用乙酸乙脂∶甲醇∶水∶氨水 =5∶9∶1∶1.5混合液作为硅胶薄板展开剂时 ,可将不同聚合度的甲壳胺寡糖很好地分开 ,且层析重现性高 ,糖残基数目和Rf值具有很好的线性关系。 2种分析方法结合可以作为 1种可靠的甲壳胺寡糖分析方法。
The standard chitosan-oligosaccharides were separated by using a novel SUGAR-KS-802 column and TLC methods. The optimal solvent system and HPLC conditions were found. Chitosan-oligosaccharides can be easily separated by using the SUGAR-KS-802 column under the condition of 0.001mol/L NaOH at 0.2mL/min, and chitosan-oligosaccharides ranging from dp1 to dp6 can be clearly separated with TLC using ethyl acetate/methano/water/ammonia water in 5/9/1/1.5 as developing system.The combination of HPLC and TLC analysis is a reliable method for the analysis and separation of chitosan-oligosaccharides.
