摘要
目的:探讨急性白血病(AL)患者多药耐药基因(mdr1)和谷胱甘肽蛳S蛳转移酶pi(GSTpi)的表达及临床意义。方法:采用反转录蛳多聚酶链(RT蛳PCR)技术检测了56 例AL患者mdr1和GSTpi的表达情况。结果:mdr1基因阳性表达率在20 例初治和17 例完全缓解(CR)的病例中分别为25 %和17.65 %,显著低于难治/复发组(78.95 %,P<0.01)。GSTpi 在56 例AL患者中有9 例表达阳性,其中初治组3 例,难治/复发组6 例;与mdr1均阳性表达有7 例。初治组mdr1基因阳性与基因阴性患者的CR率分别为20 %和86.67 %,两者间差异存在显著性(P<0.05)。结论:mdr1基因的表达是判断疗效及预后的一个重要指标。GSTpi与mdr1基因均表达阳性对于疗效和预后判断可能更具价值。
Obejective:To detect the expression of multidrug resistance(mdr1)gene and glutathione-s-transferase pi(GSTpi)in acute leukemia(AL)and its clinical significance.Methods:The expression of mdr1gene and GSTpi were measured in56patients with acute leukemia by RT蛳PCR technique.Results:The mdr1positive rate in20untreated patients and in17cases at complete remission(CR)phase were25% and17.65%,respectively,which were significantly lower than those in refractory-relapsed group(78.95%)(P<0.01).We also found that9in56AL patients were GSTpi positive(3in untreated group,6in refractory-relapsed group).The co-expression of GSTpi and mdr1was in7cases.CR rate in the mdr1positive group was lower than that in the mdr1negative group(P<0.01).Conclusion:The detection of mdr1expression is helpful in management of AL patients.The co-expression of mdr1and GSTpi may be more valuable for the judgement of therapeutic effect and prognosis in these patients.
