羟基喜树碱对人舌癌细胞的抑制作用及端粒酶活性的影响 被引量:2

摘要 目的 研究化疗药物羟基喜树碱对人舌癌Tca8113细胞的抑制作用和对端粒酶活性的影响 ,初步探讨端粒酶作为化疗敏感性指标的可行性。方法 应用MTT法、双层琼脂培养法、透射电镜观察、流式细胞术 ,TRAP -PCR -ELISA法研究。结果 羟基喜树碱对人舌癌Tca8113细胞有明显的抑制作用且呈浓度依赖性 ,细胞超微结构有明显改变 ,细胞周期发生明显变化 (S期细胞由 2 3.9%变为 39.5 % ,G0 /G1期细胞由72 .4 %变为 39.0 % ) ,抑制端粒酶活性且有一定的时间依赖性 (用 10 9ng/ml羟基喜树碱处理Tca8113舌癌细胞后 2 4 ,4 8,72和 96h端粒酶活性分别为 (0 .5 7± 0 .0 7) ,(0 .35± 0 .0 2 ) ,(0 .18± 0 .0 4 ) ,(阴性 )。结论 羟基喜树碱可以明显抑制Tca8113细胞的增值活性及其转移复发倾向 ,还可作用于线粒体 ,在细胞周期改变的同时抑制端粒酶活性。 Objective:To investigate the inhibitory effect of Hydroxycamptothecin(HCPT) on cell activity and telomerase activity in Tca8113 . Methods: We used methods of MTT assay in vitro, clonogenic assay, transmission electron microscope observation, flow cytometry and TRAP-PCR-ELISA in those untreated and treated with HCPT. Results:Telomerase activity was significantly inhibited when treated with HCPT (cell at S stage inaresed after 23.9% to 39.5%,at the stage of G 0/G 1,it decreased from 72.4% to 39.0%), and the effect of inhibition was time-dependant,By applying 109 ng/ml HCPT to the cell of togue cancer,the telomerase activity at 24,48,72 and 9 h was respectively (0.57±0.07),(0.35±0.02),(0.18±0.04),negtive .Conclusion:The results indicated that HCPT can effectively inhibit telomerase and cell activity of Tca8113 cells.meanwhile it can change the cell cycle.
