摘要
目的 :研究 5 -氟尿嘧啶对人舌癌Tca8113细胞的抑制作用和对端粒酶活性的影响 ,进一步探讨它的抗癌机理。方法 :应用MTT法、双层琼脂培养法、倒置显微镜观察、HE染色观察、透射电镜观察研究细胞用药前后的生长增殖活性和大体及超微结构变化 ,用免疫组化法、TRAP -PCR -ELISA法研究细胞用药前后部分癌基因表达及端粒酶活性的变化。结果 :5 -氟尿嘧啶对人舌癌Tca8113细胞有明显的抑制作用且呈浓度依赖性 ,对细胞大体形态和超微结构有明显的改变 ,下调c -myc、bac - 2的表达 ,抑制端粒酶活性且有一定的时间依赖性 (用 876ng/ml5 -氟尿嘧啶处理舌癌细胞后 12h、2 4h、4 8h、72h、96h端粒酶活性分别为 0 .76± 0 .0 3、0 .4 5± 0 .0 6、0 .32± 0 .0 5、0 .14± 0 .0 2 ,阴性 )。结论 :5 -氟尿嘧啶可以明显抑制Tca8113细胞的增殖活性及其转移复发倾向 ,还可以作用于线粒体 ,在下调c -myc、bac -
Objective:To study the effect of inhibit of cell activi ty and telomerase activity in Tca8113 treated by 5-fluorouracil.Methods :Using the MTT assay in vitor,clonogenic assay,HE staining,transmissi on electron microscope observation, immunohistochemical staining and TRAP-PCR-EL ISA in untreated and treated with 5-fluorouraci.Results:Te lomerase activity was significantly inhibited when treated with 5-fluorouracil, and the effect of inhibition was time-dependant.Cell activity of Tca8113 was sig nificantly inhibited, morphology of cell was changed, the levels of c-myc、bac- 2 was decreased compared with untreated Tca8113 cells.Conclusions:The results indicated that 5-fluorouracil can effectively inhibit telome rase and cell activity of Tca8113 cells.It can reduce the levels of c-myc、bac- 2 at the same time.
