摘要
[目的]研究CYP2E1RsaⅠ基因型、生活习惯因素相互作用与胃癌易感性的关系。[方法]在消化道癌高发区淮安市进行了病例对照研究(胃癌145例,人群对照229例),调查研究对象的烟酒茶嗜好及饮食习惯,以PCR扩增、RsaⅠ内切酶消化,分析研究对象的CYP2E1的基因型。[结果]胃癌组与对照组的CYP2E1RsaⅠc1/c2或c2/c2基因型频率无显著差异(41.4%∶38.9%)。吸烟和常吃腌菜与CYP2E1c1/c2或c2/c2基因型在增加胃癌发生的危险性中有相乘效应。饮酒和不饮茶的习惯增加胃癌发生的危险性,但饮酒或不饮茶的这种作用不受CYP2E1基因型的影响。[结论]CYP2E1RsaⅠ基因多态与吸烟及常吃腌菜之间的基因—环境交互作用影响个体对胃癌的易感性。
To evaluate interaction between polymorphisms in CYP2E1RsaⅠand lifestyle with risk to develop of stomach cancer .A case-control study was conducted in145cases of stomach cancer and22 9population-based controls in Huai'an city of Jiangsu Province,PRC.The epidemio logical data were collected,and DNA of peripher-al blood leukocytes was obtaine d from all of the subjects.CYP2E1genotypes were identified by PCR amplification fol-lowed by digestion with RsaⅠ.No significant difference was observ ed in the frequency of CYP2E1RsaⅠc1/c2and c2/c2genotypes between controls(38.9% )and stomach cancer cases(41.4%).Individuals who had CYP2E1RsaⅠc1/c2or c2/c2gen otype and with smoking habit were at greatly higher risk of developing stomach c ancer(adjusted OR=2.91,95%CI:1.27~6.69)compared with those who had c1/c1genotyp e and no smoking habit.Individuals who had CYP2E1RsaⅠc1/c2or c2/c2genotype and who had frequenly pickled vegetables intake increased risk of developing stomach cancer(adjusted OR=2.08,95%CI:1.00~4.32)compared with those who had c1/c1genot ype and who had less pickled vegetables intake.Habits of alcohol drinking or no tea drinking significantly increased the OR for stomach cancer.This phenomena wa s not influenced by CYP2E1RsaⅠgenotype.[Conclusions]In the present study the re sults suggested that gene-environment interaction between the CYP2E1polymorphis m and smoking,intake of pickled vegetable may in-fluence susceptibility of stom ach cancer.
