摘要
目的 :研究纤维素酶对壳聚糖的降解作用以及初步探讨壳聚糖水解反应的动力学。方法 :测定反应液的粘度并用高压液像色谱 (HPL C)分析壳聚糖水解的产物。结果 :在 18℃下水解 180 min后 ,壳聚糖溶液的粘度下降百分率为62 .4 1%。水解的产物有氨基葡萄糖和它的二糖、三糖和四糖。水解反应随酶浓度的升高而加快 ,纤维素酶作用的最适温度为 4 0℃。结论
Objective:To study the effect of cellulase on chitosan hydrolysis and its hydrolytic kinetics.Methods:The viscosity and hydrolytic products of chitosan solution were determined by viscosimeter and high performance liquid chromatography (HPLC),respectively.Results:The viscosity of chitosan solution decreased by 62.41% with enzymic hydrolysis for 180 minutes at 18℃.The hydrolysates included chitosamine,chitotriose,chitobiose and so on.The speed of enzymic hydrolysis increased with the concentration of cellulase,and the optimal temperature was 40℃.Conclusion:Cellulase can effectively hydrolyze chitosan.
