摘要
进行了温度对巴西橡胶树(Heveabrasiliensis)微体繁殖影响的研究。以巴西橡胶树的腋芽和顶芽为外植体,分别在26,28和30℃3种温度下进行组织培养,定期观测萌生枝的高度、直径、数量等多项指标。结果表明:在培养第50天时,腋芽繁殖系数从26℃的2.6提高到28℃和30℃的4.6,提高了76.9%,在同样情况下,28~30℃的顶芽繁殖系数比26℃的提高50%左右;28℃的萌生枝直径略大于30℃和26℃的;30℃和28℃的萌生枝和萌动芽数量大于26℃的;30℃的枯顶数量略大于28℃和26℃的。综合指标说明:培养温度以28℃最佳,继代1次时间不宜超过50d。
Axillary buds and apical buds of rubber trees(Hevea brasiliensis)were used as explants and cultured in vitro at 26°C,28°C and30°C.The propagation coefficient of the axillary buds at 28°C and30°C was4.5,70%higher than that at26°C(2.6).The propagation coefficient of the apical buds at28°C and30°C is50%higher than that at26°C.The diameter of the imps cultured at28°C was a bit bigger than that at30°C and26°C.The quantity of the imps was more at30°C and28°C than at26°C.The new shoots suffered apical dieback slightly more at30°C than at28°C and26°C.The data showed the best cultural temperature was at28°C.The growth peak of new shoots appeared in20to30days.The new shoots grew slowly after the50th day at26°C and28°C and the40th day at30°C.This indicates that the new shoots should not be subcultured for more than50days.
