摘要
目的 研究三角帆蚌多糖的抗肿瘤活性。方法 给荷HepA腹水瘤小鼠胃饲三角帆蚌多糖 (每日剂量 5 0 ,2 5 0和5 0 0mg/kg) 8d ,检测抑瘤率、胸腺指数和脾指数 ;采用3H TdR掺入法 ,检测三角帆蚌多糖对HepA腹水瘤细胞DNA合成的影响。结果 在上述三种试验剂量 ,三角帆蚌多糖对小鼠HepA腹水瘤的抑制率分别为 3 4.9%、48.6%和 5 1.0 % ;与对照组比较 ,试验组的胸腺指数分别提高 5 5 .6% ,61.1%和 77.8% ,脾指数分别提高 8.2 % ,12 .3 %和 16.3 % ;在离体细胞培养试验中仅高浓度组 (10 0 0 μg/mL)三角帆蚌多糖对HepA瘤细胞DNA合成和细胞增殖有显著的抑制作用 (P <0 .0 5 )。 结论 三角帆蚌多糖有一定的抗肿瘤活性 ,其作用机制可能与增强胸腺免疫功能有关。
OBJECTIVE To study the inhibitive effects of polysaccharide from Hyriopsis cumingii(HCP) on HepA(Hepa tuma) tumor proliferation. METHOD Three groups of mice bearing HepA were treated with the HCP(at dose of 50?250?500mg/kg per day, respectively) by ig for 8 days, and the tumor growth rate, the thymus gland index and the spleen index were measured. In another experiment, the HepA cells cultured in medium containing the HCP(250,500 and 1000μg/mL) and 3H TdR to test the inhibitive effect of HCP on DNA synthesis of HepA cells were detected.RESULTS In the mice treated with HCP of the doses mentioned above, the tumor growth was inhibited by 34.9%,48.6%,51.1%, with the thymus gland index enhanced by was 55.6%,61.1%and 77.8%,and the spleen index increased by 8.2%,12.3% and 16.3%, compared with respective control. In the in vitro experiment, at a dose of 1 000ug/mL, the HCP significantly suppressed the DNA synthesis in cultured HepA cells. CONCLUSION The HCP inhibited the proliferation of HepA tumor. The mechanism for the the anticarinogenic effect was related to the up regulating the immunologic function of thymus gland.
