文蛤抗癌活性多肽的生理活性研究 被引量:16

Studies on the Physiological Activity of a Natural Peptide from Clam (Meretrix meretrix Linnaeus )

摘要 以新鲜文蛤为材料,经有机溶剂萃取,分子筛凝胶层析,得到一种具有抗癌生物活性的文蛤多肽,SDS PAGE电泳测得该多肽的分子量约为18kD.用该多肽处理人胃腺癌细胞BGC 823,结果表明,当多肽浓度为10μg/mL,细胞生长的抑制率达到50%.此外,探讨了该活性多肽在体外对几种与肿瘤代谢相关的酶的效应,实验结果显示,该活性多肽能够激活碱性磷酸酶和超氧化物歧化酶,当其浓度为10μg/mL时,可以使这两种酶活力分别提高150%和93%;该活性多肽对酪氨酸酶则是起到了抑制的效应,测定导致酶活力下降50%所需的效应物浓度(IC50)为1.50μg/mL,其抑制类型为混合型抑制,其抑制常数KI和KIS分别为4.37mg/mL和6.09mg/mL. A kind of peptide (M_2) was purified from greenness clam (Meretrix meretrix Linnaeus) by the method of alcoholic extraction, gel filtration and so on. The final preparation was homogeneous on SDS-PAGE. The molecular weight was determined to be 18.4 kD by the method of SDS-PAGE electrophoresis. According to the research, this kind of peptide has the inhibitory effect on the growth of human gastric cancer cells BGC-823. The inhibitory rate was nearly 50% when they were treated by M_2 at the concentration of 10 μg/ mL. A survey to the effects of M_2 on several enzymes which were relative to the metabolizing ability of tumors showed that this peptide could activate alkaline phosphatase and superoxidedismutase, but inhibit the of tyrosinase. When the concentration of M_2 was 10 μg/ mL, the peptide can enhance the activity of ALP and SOD by 150% and 93%, respectively. The inhibitory effect on the tyrosinase activity was further studied and the results showed that when the concentration of M_2 came to 1.50 μg/ mL, the inhibitory rate of tyrosinases activity was fell 50%. The inhibitory mechanism of the peptide on tyrosinase was studied and the results showed that it is a kind of mixed-type inhibitor to tyrosinase. The Inhibition constantK_I andK_(IS) were determined to be 4.37 μg/ mL and 6.09 μg/ mL, respectively.
