摘要
优化超声波浸提蒲公英根甾醇的工艺条件。试验控制1.00 g样品用20 mL乙醇在超声波清洗机里进行浸提,设计正交试验和响应面试验研究甾醇提取率随着乙醇浓度、超声提取时间、超声提取温度三个因素水平变化而变化的规律。正交优化分析得出最佳提取工艺下,能提出甾醇含量为46.01 mg/g,此时乙醇浓度为80%,提取时间为20 min,提取温度为60℃。响应面分析结果为乙醇浓度87.42%、提取时间10.83 min、提取温度59.97℃,这时每克蒲公英根提取甾醇的含量最高为67.26 mg。该结果表明响应面法工艺条件下得到的蒲公英根甾醇提取率高于正交试验所得。
Optimization of process conditions for ultrasonic extraction of Taraxacum mongolicum root sterols.Experimentally controlled 1.00 g samples were extracted with 20 mL of ethanol in an ultrasonic cleaner.Orthogonal and response surface tests were designed to investigate the pattern of sterol extraction rate with the changes in the levels of three factors:ethanol concentration,ultrasonic extraction time,and ultrasonic extraction temperature.The orthogonal optimization analysis concluded that under the best extraction process,the sterol content was 46.01 mg/g,and the ethanol concentration was 80%,the extraction time was 20 min,and the extraction temperature was 60℃.The results of the response surface analysis were 87.42%ethanol concentration,10.83 min extraction time and 59.97℃extraction temperature,and the highest sterol content of 67.26 mg per gram of T.mongolicum root extracted.This result suggested that the sterol extraction rate of T.mongolicum root obtained under the process conditions of response surface method was higher than that obtained from the orthogonal test.
