摘要
本文研究筛选出了薄层色谱——氯胺T浓硫酸比色法,它可以用于油脂及油脂精炼下脚无甲基甾醇的定量分析。无甲基甾醇经簿层分离后在紫外灯照射下呈现紫颜色谱带(Rf值约0.38),与氯胺T浓硫酸反应能生成稳定的棕黄色物质,在波长400nm处具有最大吸收值。该方法和内标法相比具有溶剂少、快速、精度高、重现性好,操作较简便等优点,至今在国内外文献中尚未有过报道。
A thin layer chromatography (TLC) -colorimetric method using chloramine T and cone sulfuric acid reagent was screened out for quantitative analysis of demethlyate sterel in vegetable oil and refuse of refined oil. Demethlyate sterol was separated with thin layer chromatography. The yellowish-brown colored product of reaction of sterol with ehloramine T in cone sulfuric acid had its maxium absorption at 400 nm. Comparing with the internal standard method, it has the advantages of little solvents, higher precision, fine reproducibility and simpler procedure, etc.
