摘要
目的建立蒲公英中菊苣酸、绿原酸、单咖啡酰酒石酸、异绿原酸A、木犀草苷、隐绿原酸6个成分的超高效液相色谱(UPLC)含量测定方法。方法采用Acquity UPLC BEH-C18(100 mm×2.1 mm,1.7μm)色谱柱,0.1%的磷酸水溶液-乙腈为流动相,梯度洗脱,流速为0.4 mL/min,检测波长为327 nm,柱温为35℃。结果6个成分线性关系良好(r>0.9990),精密度均小于3%,平均回收率在99.22%~107.16%之间。不同来源的蒲公英中6个活性成分含量差异较大。蒲公英叶片中活性成分(尤其菊苣酸)显著高于根,根中基本不含隐绿原酸和木犀草苷。结论该方法准确、简便、重复性好,可用于蒲公英的质量控制和评价,同时也为蒲公英不同药用部位开发提供参考。
Objectivee To establish an ultra-high performance liquid chromatography(UPLC)method for the determination of the contents of 6 components in Taraxacum mongolicum,including chicoric acid,chlorogenic acid,caftaric acid,isochlorogenic acid A,luteolin-7-O-glucoside and cryptochlorogenic acid.Methods An Acquity UPLC BEH-Cis chromatographic column(100 mm×2.1 mm,1.7μm)was used,with a mobile phase consisting of a 0.1%phosphoric acid solution and acetonitrile in gradient elution mode.The flow rate was 0.4 mL/min,the detection wavelength was 327 nm,and the column temperature was 35℃.Results The linear relationships of the 6 components were good(r>0.9990),with precision values all less than 3%,and the average recovery rates ranging from 99.22%to 107.16%.Significant differences were observed in the contents of the 6 components among T.mongolicum samples from different sources.The active components(especially chicoric acid)in leaves were significantly higher than those in the roots,which contained almost no luteolin-7-O-glucoside and cryptochlorogenic acid.Conclusion The method is accurate,simple,and has good reproducibility,making it suitable for the quality control and evaluation of T.mongolicum.The study on the content differences between roots and leaves can provide reference for the development of different medicinal parts of T.mongolicum.
