天宁饮对硝酸甘油所致偏头痛大鼠行为症状的影响 被引量:4

Efects of Tianning Decoction on Behavior and Symptoms of Nitroglycerin Rat Migraine Model

摘要 目的:研究天宁饮对硝酸甘油所致偏头痛大鼠行为症状及三叉神经核尾部c-fos蛋白表达的影响,从动物实验角度证实天宁饮的疗效及其作用机理。方法:将60只雌雄各半SD大鼠随机分为空白对照组、偏头痛模型组、阳性对照组(盐酸氟桂利嗪组)、天宁饮高、中、低剂量组,每组10只。除空白对照组外,其他组采用皮下注射硝酸甘油(10 mg/kg)复制偏头痛大鼠模型,造模后30 min分别给予蒸馏水及药物灌胃,观察各组大鼠耳红开始、结束时间及不同时间挠头、爬笼次数;同时观察模型大鼠三叉神经核尾部c-fos蛋白表达情况。结果:与空白组比较,模型组出现耳红现象,挠头、爬笼次数显著性增加(P<0.01),三叉神经核尾部c-fos蛋白表达亦显著性增加(P<0.01);与模型组比较,阳性对照组(盐酸氟桂利嗪组)、天宁饮高、中、低剂量组均可明显缩短模型大鼠耳红时间,抑制模型大鼠挠头、爬笼次数(P<0.05),显著性抑制模型大鼠三叉神经核尾部c-fos蛋白表达(P<0.01)。结论:天宁饮可有效治疗硝酸甘油型所致大鼠偏头痛,其作用机制与抑制模型大鼠三叉神经核尾部c-fos蛋白表达有关。 Objective: To study the effects of Tianning Decoction on behavior and symptoms of nitroglycerin migraine rat model and the trigeminal nucleus c - fos protein expression and to prove Tianning Decoction curative effect and its mechanism of action by animal experiments. Methods : 30 male and 30 female SD rats were randomly divided into blank control group, model group, positive control group (flunarizine hydrochloride) high, medium and low dose groups and each group had 10. Except the blank control group,the other groups made models of migraine rats by subcutaneous injection of nitroglycerin injection (10 mg,/kg) . After 30 minutes,rats were fed with Tianning Decoction and distilled water by groups. Observe time of ears starting turning red and stopping of each group and times of scratching his head and times of climbing cage. At the same time, observe the change of c - fos protein expression of trigeminal nucleus tail of migraine rats. Results: Compared with blank group, time of ears of rats of model group appearing ear red, times of scratching his head and the times of climbing cage were significantly increased (P 〈 0.01 ) and the trigeminal nucleus tail c - fos protein expression was also significantly increased ( P 〈 0.01 ). Compared with model group,positive control group ( flunarizine hydrochloride), high, medium and low dose groups of Tianning Decoction can obviously shorten the model rat ear red time, inhibit the times of model rats scratching his head and the times of climbing cage (P 〈 0. 05 ) and c - fos protein expression of the trigeminal nucleus tail was significantly inhibited (P 〈 0.01 ). Conclusion: Tianning Decoction can be effective in the treatment of nitroglycerin migraine rats and its mechanism is related to inhibiting c - fos protein expression of trigeminal nucleus of rats tail.
