摘要 目的研究胰岛素样生长因子1(IGF-1)对神经元p75受体表达的影响,探讨IGF-1在神经特异性再生中的作用机制。方法在体外培养神经元培养基中加入不同浓度的重组IGF-1,用粘附式细胞仪检测神经元表面神经生长因子p75受体的表达。结果IGF-1作用后神经元p75受体的表达增加,细胞表面p75受体的增加与IGF-1的浓度在一定范围内呈现量效关系。结论IGF-1在体外培养条件下可增加神经细胞表面p75受体的表达,IGF-1的神经营养作用机制可能与p75受体表达上调有关。 Objective To study the effects insulin-like growth factor1(IGF-1)on the expression of nerve growth fac tor low affinity receptor p75in neurons and investigate the mechanismof IGF-1acting during nerve specific regeneration.Methods The primary cultured neurons in vitr o were incubated with IGF-1of different co ncentration.The influence of IGF-1on expression of p75on neurons was me asured by confocal microscopy.Results The expression of p75in neurons was r emarkably increased with the use of I GF-1.There was dose-dependent effe ct relationship between IGF-1concentration and the expression of p75at the concentration of 5~80ng /ml.Conclusion IGF-1has the abilities of increasin g expression of p75on cultured neurons in vitro.The mechanism of IGF-1promoting neuron s survival may due to the up-grade regulation of p75.
