摘要
为了证实生长因子对体外培养成熟听神经元的影响,应用无血清培养技术和细胞学定量方法,观察了神经生长因子(NGF)、硷基成纤维细胞生长因子(bFGF)对生后10~16日龄白莱亨鸡初级听神经元的体外存活及其轴突再生的生物学作用。结果表明,出生后鸡的听神经元在生长因子作用下仍可在体外存活。早期生长细胞以非神经元成份(Schwann细胞,成纤维细胞)为主,24小时可见听神经元突起(双极或单极)形成。培养48~72小时轴突生长旺盛。实验表明,外源性NGF与bFGF对幼鸡听神经元具有直接的营养作用。两者在听神经元的体外存活与轴突再生过程中呈现出协同作用。上述生物效应在体外条件下存在浓度依赖性与饱和特点。
Toconfirmthebiologicalefectsofpeptidegrowthfactorsonauditoryneuron(AN)invitro,theinfluenceofNGFandbFGFonthesurvivalstatusandtheneuriteregrowthofdissociat-edprimaryANinpostnatalchickswasstudiedwithserum-freeculturetechniqueandcytologicquantitativemethod.Theresultsdemonstratedthatthegrowthofnon-neuronelementssuchasSchwann′scelswaspredominantintheearlierstageofculture.Theneuriteregrowth(bipolarityorunipolarity)ofANwasobservedat24hoursafterseeding.Thegrowthrateincreasedduring48-72hoursinthesurvivingAN.Thesefindingsindicatedthatbothgrowthfactorsexerteddirectneu-ronotrophicefectondissociatedANinvitro.NGFandbFGFactedincooperationtopromotetheax-onregenerationofAN.Theconcentration-dependenteffectofNGFonneuriteregrowthandneuronsurvivalwasfoundinthepresentstudy.ThebiologicalmechanismofNGFandbFGFonANinvitrohasbeendiscussed.
