摘要
目的:研究大鼠慢性压迫性脊髓损伤及减压后骨骼肌萎缩与细胞凋亡的关系以及凋亡相关基因P53的表达意义。方法:45只SD大鼠分成3组,压迫组15只,作成轻、中和重度慢性脊髓压迫损伤;减压组25只,将重度压迫组的压迫装置取出;正常对照组5只,不做任何处理。应用苏木精-伊红染色、终末脱氧核苷酰转移酶介导的原位末端标记法和原位杂交技术研究慢性脊髓压迫及减压后肌细胞萎缩、凋亡的特点。结果:随着压迫程度的增加肌细胞凋亡数量增加,P53mRNA表达增高;减压后肌细胞凋亡数量明显减少,P53mRNA表达减少。结论:慢性脊髓压迫后肌细胞出现凋亡,骨骼肌细胞的凋亡是肌肉失神经萎缩的机制之一,减压可通过抑制细胞凋亡阻止肌肉萎缩。
AIM:To study the relationship between skeletal muscular atrophy and apoptosis after chronic compressive spinal cord injury and decompression, and the signific ance of apoptosis-related genes of P53. METHODS:Forty-five SD rats were divided into three groups: group A (mild, mod erate and severe chronic degree compressive injury of spinal cord) of 15 samples , group B(decompression) of 25 samples and group C(normal control) of 5 samples. Hematoxylin and eosin(HE) staining and TUNEL and hybridization in situ were use d to investigate the apoptosis and atrophy of muscular. RESULTS:With increased of compression degree, the number of apoptosis nuclear and the expression of P53 mRNA increased. The number of apoptosis nuclear and th e expression of P53 mRNA decreased after decompression of spinal cord.
