摘要
目的:探讨右归丸对实验性自身免疫性脑脊髓炎小鼠(experimental autoimmune encephalomyelitis,EAE)IL-23/IL-17炎性轴的影响。方法:建立髓鞘少突胶质细胞糖蛋白35-55(myelin oligodendrocyte glycoprotein 35-55,MOG35-55)诱导C57BL/6小鼠EAE模型,随机分为模型组、泼尼松组,右归丸组,并设正常对照组。观察小鼠的发病情况、神经功能评分及体重变化,HE染色观察CNS病理损伤程度,实时荧光定量PCR法检测小鼠脑组织IL-23 p19、IL-17 mRNA表达。结果:与正常对照组比较,模型组脑组织IL-23 p19、IL-17mRNA表达显著升高,泼尼松及右归丸组小鼠脑组织IL-23 p19、IL-17的mRNA表达比模型组低(P<0.01)。结论:右归丸能够抑制IL-23/IL-17炎性轴而起到抗炎治疗作用。
Objective:To investigate the effects of Yougui Pill on IL -23/IL - 17 axis of mice with acute experimental autoim- mune encephalomyelitis (EAE). Methods :EAE model was induced in C57BL/6 mice,and was treated with Yougui Pill (Yougui Pill group) ,prednisone (prednisone group) and saline( EAE group). Normal mice were selected as control group. Levels of IL- 23p19 ,IL- 17 mRNA expression in brain tissue were detected by real time- PCR. Results:IL- 23p19, IL- 17 mRNA levels in brain tissue were significantly decreased in Yougui Pill and prednisone treated EAE mice than those in EAE mice (P 〈 0.01 ). Conclusion:Yougui Pill may exert anti -inflammatory effect through inhibiting IL- 23/IL- 17 axis.
