大鼠坐骨神经移植的形态及电生理研究

摘要 目的:探讨异体坐骨神经移植后神经纤维再生。方法:大鼠38只,在手术显微镜下将右侧坐骨神经切除1cm,然后将1cm异体或自体右侧坐骨神经移植于神经缺损处。术后2、4、8、12周进行形态学观察及神经电生理学检查。结果:术后4周再生神经纤维通过近侧吻合口进入移植神经,术后8周通过远侧吻合口进入受体坐骨神经。术后12周异体组与自体组再生神经纤维髓鞘的厚度、轴索的形状及神经电生理功能无明显差异。结论:免疫排斥反应后溃变的异体移植神经可作为桥梁和支架,使再生的神经纤维通过移植神经到达靶组织,在一定的程度上恢复其功能。 Objective: To study the nerve fiber regeneration after allogenic peripheral nerve transplantation. Methods: 1 cm allogenic or autogenous right sciatic nerve was grafted to the site where 1 cm nerve of right sciatic nerve was cut. Morphologic observation and electrophysiological exami-?222 ?nations were taken 2,4,8 and 12 weeks after operation. Results; Regenerating nerve fibers passed through the proximal anastomotic juncture and entered the grafted nerve 4 weeks after the operation, and also into the distal part of the recipient sciatic nerve 8 weeks after the operation. The thickness of the myelin sheath, ultrastructure of axon from the regenerating nerve fibers and results of electrophysiological examinations were similar between allogenic and autogenous groups. Conclusion: The degenerated nerve fibers may serve as a framework or bridge for the regeneration of nerve fibers after immune rejection. Those nerve fibers are able to pass through the grafted nerve and reinnervate the target tissue and restore their functions in some degree.
