摘要
目的研究3-溴丙酮酸对SWⅢ-C细胞增殖和凋亡的影响。方法利用透射电镜、MTT法、流式细胞术、免疫组化等方法检测3-溴丙酮酸作用于SWⅢ-C细胞后,其细胞形态、超微结构的改变;细胞增殖、细胞周期分布和细胞凋亡率的变化以及Bcl-2、c-myc和mtp53蛋白表达的变化。结果 3-溴丙酮酸可抑制SWⅢ-C细胞的增殖,浓度为25μg·mL-1时抑制作用最明显;3-溴丙酮酸可有效诱导SWⅢ-C细胞凋亡,浓度为25μg·mL-1效果最明显;3-溴丙酮酸可下调SWⅢ-C细胞Bcl-2、c-myc和mtp53蛋白的表达。结论 3-溴丙酮酸可抑制SWⅢ-C细胞增殖并诱导其凋亡,其机制可能与下调Bcl-2、c-myc和mtp53的表达有关。
Objective To investigate the effects of 3-bromine pyruvate on the proliferation and apoptosis of SWIII-C cells. Methods The methods with transmission electron microscope, MTT, Flow Cytometry, immunohistochemistry and so on were used to detect the changes of cellular morphology and ultramicrostmcture, changes of cell proliferation, cycle distribution and apoptosis rate, and changes of Bcl-2, c-myc and mtp53 protein expression of SWIII-C cells after giving 3-bromine pymvate. Results 3-bromine pymvate has the obvious inhibition effect on SWIII-C cell. Especially when its concentration reached up to 25 pg.mL-~, it achieved the best inhibition effect. The protein expression ofBcl-2, c-myc and mtp53 gene of SWIII-C cells might be regulated down by 3-bromine pyruvate. Conclusion 3-bromine pymvate may inhibit the proliferation and induce the apoptosis of SWIII-C cells, and the mechanism may associate with the down-regulation of expression ofBcl-2, c-myc and mtp53.
