摘要 目的:探讨β-catenin(β-连环素)在大肠癌的表达及其与癌基因cyclinD1和C-myc的关系。 方法:应用免疫组化Powervision^(TM)二步法,检测25名正常人大肠黏膜,42例大肠腺瘤和58例大肠癌组织β-catenin,cyclinD1和C-myc的表达情况。 结果:正常大肠黏膜β-catenin胞膜阳性表达,cyclinD1和C-myc阴性。大肠腺瘤和大肠癌组织β-catenin呈胞质和/或胞核异位表达,大肠癌异位表达率为65.5%,显著高于大肠腺瘤(42.9%,P<0.05)。大肠癌cyclinD1和C-myc阳性率分别为74.1%、70.7%,均显著高于大肠腺瘤(45.2%,33.3%;P均<0.01)。大肠癌中β-catenin与cyclinD1和C-myc表达呈明显正相关(r=0.32,0.57;P<0.05,P<0.01)。 结论:β-catenin异位表达可能是原癌基因cyclinD1和C-myc激活的原因之一,并在大肠癌发生过程中可能起重要作用。 AIM: To detect the expression of β-catenin and to explore its correlation with the expression of oncogene cyclin D1 and c-myc in the colorectal carcinoma.METHODS: Expression of β-catenin, cyclin Dl and c-myc was examined by PowerVision?immunohistochemistry in 25 cases of normal colonic mucosa, 42 colorectal adenoma and 58 colorectal carcinoma.RESULTS: In normal colonic mucosa, β-catenin was positive in the cell membranes of epithelial cells, while cyclin D1 and c-myc were negative. The cytoplasmic and nuclearβ-catenin expression were found in the colorectal adenoma and carcinoma. The cytoplasmic and nuclear β-catenin expression rate was 65.5 % in colorectal carcinoma, which was significantly higher than that of colorectal adenoma (42. 9 %, P < 0.05). The positive expression rate of cyclin D1 and c-myc in the colorectal carcinoma were 74.1 % and 70.7 %, respectively, and both of the rates were significantly higher than that of colorectal adenoma (45.2 %, 33.3 %, P < 0. 01, for all). The cytoplasmic and nuclearβ-catenin expression rate was significantly correlated with the positive expression rate of cyclin D1 and c-myc in the colorectal carcinoma (r= 0.32,0.57; P<0.05, P<0.01). CONCLUSION :Cytoplasmic and nuclear β-catenin expression may activate the expression of oncogene cyclin D1 and c-myc, which plays a pivotal role in the carcinogenesis and progression of colorectal carcinoma.
