大孔吸附树脂纯化甘草总皂苷 被引量:5

Purification of total saponin of Licorice root by macroporous adsorptive resin

摘要 目的:研究甘草总皂苷的大孔吸附树脂分离纯化工艺。方法:以紫外分光光度法测定3种树脂对甘草总皂苷的吸附率、解吸率,以甘草总皂苷的吸附率、洗脱率、比吸附量、比洗脱量为考察指标综合评价,对吸附、纯化等工艺参数进行考察。结果:将5mg·mL-1(甘草总皂苷含量)的提取液pH调为7.0,以2BV·h-1的流速上样于D-101大孔吸附树脂柱,用2BV水洗净,pH为8.0~9.0的50%乙醇以2BV·h-1的速度洗脱,收集4BV的洗脱液洗脱效果较好。结论:甘草总皂苷部位可使甘草总皂苷的收率和质量分数达到较满意效果。 OBJECTIVE To study purification technology of total saponin of licorice root by macroporous adsorptive resin. METHODS Adsorbtion ratio and eluation ratio of 3 kinds resin to total saponin of licorice root were detected by UV. Processing parameters were investigated. RESULTS The best condition were:adjusting pH of the 5 mg·mL^-1 (content of total saponin of licorice root) extracting solvent to 7. 0, through D-101 macroporous adsorptive resin with 2 BV·h^-1 flow rate, and using 2 BV water clearing, eluating with pH 8.0 - 9. 0 511G alcohol with 2 BV·h^-1 rate. collecting 4 BV eluation solvent. CONCLUSION The extraction of licorice root could be conducted through D101 macroporous adsorptiveresin.
