花粉管通道介导螺旋藻总DNA导入小麦后的转基因D_1代研究 被引量:3

摘要 采用花粉管通道法将螺旋藻总DNA导入4个小麦品种9411、955159、96266、鲁麦21中,并对转基因D1代进行了研究。结果如下:与对照相比,各处理转基因种子的发芽率降低6.25%~20.21%;9411和鲁麦21的转基因D1代平均株高比对照分别降低5cm、4.6cm;在955159和鲁麦21转基因D1代中,发现麦穗的无芒变异;转基因D1代根尖细胞中均发现染色体数目的变异和染色体落后及染色体桥的类型,其变异率均在5%左右;各处理中种子粗蛋白含量表现不同程度的降低;9411和鲁麦21转基因D1代高分子量亚基发生了突变。 The total DNA of spirulina was introduced into wheat varieties 9411,955159,96266 and Lumai21 through pollen tube pathway. The D1 genration of transgenic wheat was studied and the results were as follows. Compared to the control, the germination percentage of the D1 seeds decreased by 6.25%-20.0%. The average plant height of transgenic 9411 and Lumai21 decreased 5 and 4.6 centimeter respectively. The variation of awnless ears were found in transgenic D1 of 955159 and Lumai21. The variation rates were all up to 5% of chromosome number, lagging and bridge chromosome in root tip cells. The content of crude protein decreased at different degree. The high weight subunit of glutenin varied in transgenic D1 of 9411 and Lumai21.
