花粉管通道法导入抗赤星病烤烟总DNA D_1代性状变异的研究 被引量:21

Studies on variation in characters of D_1 generation obtained by introducing total DNA of fluecured tobacco with resistance to Alternaria alternata through the pollen tube pathway

摘要 采用花粉管通道技术将抗赤星病烤烟CV87的总DNA导入感病烤烟NC89,对其D_1代植株进行了田间生长性状、抗病性和烟叶品质的检测。结果显示,柱头滴加(DZ)、切柱头滴加(QD)和子房注射(ZZ)等3种导入方法得到的D_1代植株在株高、株型、叶型、生育期等方面发生了较大的变异,变异率分别是19.10%,20%和18.20%。3类植侏中分别有9.10%,10%和6.40%的个体获得了对赤星病的抗性。在这些植株中,筛选出DZ9811和QD9812在抗病性和烟叶品质上为优良植株。 The total DNA of fluecured tobacco, CV87, with resistance to alternaria alternata, was introduced into susceptible fluecured tobacco, NC89, through pollen tube pathway. Their first generation (D_1) was analyzed in field growth traits, resistance, and quality of tobacco leaf. By the three introducing methods, ovule injecting (ZZ), stigma dropping (DZ), and cutting off stigma dropping (QD), the higher ratios of variation of plant height, plant type, leaf shape, and growth period were obtained in the D_1 generation individuals, which were 19.10%, 18.20%, and 20%, respectively. Among the plants of ZZ. QD, and DZ, there were 9.10%, 10%, and 6. 40% individuals which obtained resistance to Alternaria altrnata. DZ9811 and QD9812 are the most excellent plants among these plants with variations.
