摘要
目的探讨兔视网膜缺血一再灌注时血管内皮生长因子(vascular endothelial growth factor,VEGF)的表达,以及丹参素钠对其表达的影响。方法兔30只,随机分成3组:正常对照组3只,生理盐水对照组15只,丹参素钠组为12只,生理盐水组与丹参素钠组所有兔双眼均行视神经结扎60min诱导缺血,于缺血-再灌注开始时立即分剐给予生理盐水(1.5mL·kg^-1)、丹参素钠(0.15mg·kg^-1)静脉注射,以后每日相同时间给予相同剂量。测定生理盐水组与丹参素钠组兔视网膜缺血-再灌注0h、3h、24h、48h、168h的视网膜节细胞VEGF表达。结果生理盐水组视网膜VEGF表达自再灌注0h开始增强,至24h达到高峰,后逐渐减弱,168h时仍高于正常对照组。丹参素钠组再灌注3~48hVEGF表达与同时间段生理盐水组比较无显著性差异.168h时较生理盐水组明显减弱。结论兔视网膜缺血-再灌注期间VEGF表达增强,丹参素钠对再灌注早期VEGF的表达无明显影响,再灌注晚期可使VEGF表达减弱,对视网膜新生血管形成具有一定的预防作用。
Objective To investigate the effect of danshensu-sodium on vascular endothelial growth factor (VEGF) expression after retinal ischemiareperfusion in rabbits. Methods Thirty healthy rabbits were randomly divided into three groups including normal control group (3 rabbits), saline-treated group(15 rabbits), danshensu-sodium-treated group ( 12 rabbits). Both eyes of rabbits in the latter two groups were induced ischemia by optic nerve ligature for 60 minutes. Rabbits were respectively treated with normal saline(1.5 mL· kg^- 1 ), danshensu-sodium ( 0. 15 mg · kg^- 1 ) intravenously immediately after reperfusion,and the same dosage was given at the same time each day. VEGF expression of retinal ganglion cells was measured at 0 hour, the 3rd hour, the 24th hour, the 48th hour, the 168th hour of reperfusion after retinal ischemia. Results Retinal VEGF expression began to increase at 0 hour of reperfusion and peaked at the 24th hour, then gradually decreased. The level of VEGF expression was still higher than that of the normal control group at the 168th hour. There were no significant difference of VEGF expression between danshensu-sodium-treated group and saline-treated group at the same time during 3-48 hours of reperfusion. VEGF expression significantly decreased compared to saline-treated group at the 168th hour. Conclusion VEGF expression increases during rabbit retinal ischemia-reperfusion. VEGF expression isn' t affected by danshensu-sodium during early stage of rabbit retinal ischemiareperfusion, and decreases during later stage of reperfusion, which indicates certain preventive effect of danshensu-sodium on retinal neovascularization.
