摘要
目的在大鼠视网膜缺血-再灌注损伤模型上,研究七叶皂甙钠对视网膜缺血-再灌注损伤后视网膜内细胞凋亡的影响.方法60只大鼠随机分为对照组及七叶皂甙钠组,每组30只.通过结扎大鼠左颈总动脉1h,然后再灌注.七叶皂甙钠组腹腔注射七叶皂甙钠1.5mg/kg,对照组腹腔注射生理盐水1mL/kg,分别检测1、6、12、24、48、72h各时段大鼠视网膜内的细胞凋亡,每时段大鼠各5只.结果再灌注后,细胞凋亡主要出现于视网膜的神经节细胞层和内核层,偶尔在外核层可见凋亡细胞.再灌注后1、6、12、24、48、72h对照组细胞凋亡平均发生率分别为1.8±0.1,7.1±0.2,18.2±1.4,34.7±2.1,22.6±0.9,16.3±0.4,而在七叶皂甙钠组分别为1.7±0.2,4.2±0.6,11.9±1.1,17.8±0.9,13.5±0.7,6.8±0.4,七叶皂甙钠对细胞凋亡的平均保护效应为38.1%.结论七叶皂甙钠可以抑制再灌注后细胞凋亡的发生,对大鼠视网膜具有保护作用.
Objective To observe the effect of β -escin on retina apoptosis in rat ischemia-reperfusion injury model. Methods 60 rats were divided into control group and β-escin treated group randomly,each group 30 rats.The left common carotid artery of the rats was ligated for lh,then reperfusion followed. β-escin of 1.5mg/kg and saline of 1 mL/kg were intraperitonealy injected in β-escin treated group and control group respectively, then retinal apoptosis in 1, 6, 12, 24, 48 and 72 h was examined with each phrase for 5 rats. Rosults Apoptosis mainly occurred in retina ganglion cell layer and inner nuclear layer, occasionally in outer nuclear layer. In 1, 6, 12, 24, 48 and 72 h after reperfusion, mean retinal apoptosis rate in control group were 1.8 ± 0.1, 7.1 ±0.2, 18.2 ± 1.4, 34. 7± 2.1, 22.6 ±0.9 and 16.3 ±0.4, however, in escin group were 1.7 ± 0.2, 4.2 ±0.6, 11.9 ± 1.1, 17.8 ±0.9, 13.5 ±0.7 and 6.8 ±0.4. The mean protective effect of β-escin to apoptosis was 38.1%. Conclusion It is significant for β-escin to protect retina from ischemia-reperfusion injury.
