摘要
为了优化饲料添加剂用黄芩甙提取工艺,采用L9(34)正交试验设计方法,对浸提和酸沉工艺进行了优化试验研究,采用紫外分光光度法对提取物黄芩甙进行含量测定。结果表明:浸提较优工艺为A3B3C2,即浸提时间1.5h,浸提温度100℃,浸提pH7.0;酸沉较优工艺为A′2B′3C′1D′3,即保温时间1.0h,保温温度80℃,保温pH1.0,静置时间12h。经两次正交试验设计所得优化提取工艺可行,提取率为91.56%,含量为94.12%。
In order to optimize the extracting technology of baicalin in Scutellaria baicalensis Georgi, the orthogonal experimental design was adopted in the extraction and the acid-deposition processes, Ultra-Violet spectroscopic method was employed to determine the content of baicalin. Results proved that the best extraction technology was A_3B_3C_2, i.e. extraction time was 1.5 h, temperature was 100℃, pH value was 7.0; the best acid deposition technology was A~′_2B~′_3C~′_1D~′_3, i.e. heat preservation time was 1.0 h, temperature was 80℃, pH value was 1.0, while placement time was 12 h. The optimized technologies mentioned above were feasible. The extraction rate was 91.56%, baicalin content was 94.12% using the technologies.
