摘要
建立了一种利用反相高效液相色谱法同时测定啤酒中 9种有机酸的方法。应用反相C18色谱柱Nucleosil 10 0C18 2 5 0× 4 0mm ,以 0 1mol/LKH2 PO4(用H3 PO4调 pH3 0 )为流动相 ,紫外检测 2 15nm ,将啤酒中草酸、丙酮酸、苹果酸、α 酮戊二酸、乳酸、乙酸、柠檬酸、富马酸、琥珀酸一次性分离。除柠檬酸外各种有机酸的回收率在 85 %以上 ,变异系数 <5 %。该方法简便、快速、准确。此外 ,对影响有机酸分离的因素进行了研究 。
A reverse phase HPLC method for simultaneous determination of 9 organic acids in beer and wort has been developed.Oxalic,pyruvic,malic,ketoglutaric,lactic,acetic,citric,fumaric and succinic acids in beer and wort were separated by the use of Nucleosil 100C 18 column, 0.1 mol/L KH 2PO 4(adjusted to pH3.0 with phosphoric acid) as mobile phase and were detected by UV detector at 215nm.Recovery of most of organic acids were higher than 85% except for citric acid.Coefficients of variation is less than 5%.This method is covenient,rapid and accurate way to determinate the organic acids in beer and wort. In addition,several factors which influence the separation and determination of organic acids were studied.The concentration of organic acids in more than 10 kinds of commercial beers were detected.
