摘要 了解CML细胞和K562细胞体外诱导CML细胞特异性T细胞的TCRVβ亚家族利用、克隆性增殖和杀伤性情况。方法:采用混合淋巴细胞/白血病细胞培养法(MLTC)体外将CML细胞和K562细胞与脐血和成人外周血MNC混合培养和扩增,运用RT-PCR-基因扫描技术分析其TCRVβ亚家族的利用和克隆性特点,以及应用间接荧光单抗标记技术和LDH法分析诱导增殖的T细胞免疫表型和杀伤性。结果:CML细胞和K562细胞可以诱导出呈寡克隆或寡克隆趋势生长的Vβ亚家族T细胞,并具有识别和杀伤CML细胞的功能。结论:CML细胞和K562细胞可在体外诱导出异体CML细胞特异性CTL,该CTL可能为优势表达的Vβ亚家族克隆性T细胞。 Objective:To investigate the anti-leukemia effect,the distribution and clonal expansion of TCR Vβsubfamily T cells in T cells from cord blood and adult peripheral blood in duced by CML cells and K562cells in vitro.Methods:T cells of peripheral blood from one adult donor and3cases of cord blood were stimulated with CML cells and K562cells and far-ther am plified by liquid T cells bulk culture,to induce the CML specific cytotoxic T lymphocytes.The induced T cells were farther analyzed for the specific cytotoxicity in CML by LDH assay,the phenotype identification by indirect immunofluorescence technique and the distribution and clonal expansion of TCR Vβsubfamilies by using reverse transcriptase-polymerase chain reac tion(RT-PCR)and genescan analysis,respectively.Results:Oligoclonal and oligoclonal tendency T cells with higher specific cytoxicity from cord blood and adult peripheral blood could be in duced by stimulate with CML cells and K562cells.Conclusion:The specific cytotocity T cells for anti-CML effect could be induced by CML cells and K562cells.The induced T cells which have the ability of specific cytoxicity to the CML cells may be come from the clonal expansion TCR Vβsubfamily T cells.
