茶多酚对肺癌PG细胞生长的调控研究 被引量:4

摘要 目的 :探讨茶多酚对人 PG细胞生长的影响。方法 :采用 MTT法体外观察不同浓度茶多酚对 PG细胞的杀伤作用 ;应用激光共聚焦显微镜和流式细胞仪分析 ,检测用药后 PG细胞内 Ca2 +浓度、CD44 V6表达和细胞增殖周期分布的变化。结果 :3个浓度的茶多酚对 PG细胞均有杀伤作用 ,呈剂量依赖关系。细胞增殖受到明显抑制 ,使细胞阻滞于 G0 / G1期 ,不能进入 S期及 G2 / M期 ,细胞增殖指数明显下降。细胞内 Ca2 +浓度与对照组相比 ,逐渐上升 ;CD44 V6表达水平则呈逐渐下降。结论 :茶多酚对 PG细胞的生长具有抑制作用 ,其作用机制与改变细胞内 Ca2 +浓度和 CD44 OBJECTIVE:To explore the effects of tea polyphenols on growth of PG cell lines. METHOD:The in vitro anti proliferation effects of three different concentration of tea polyphenols on PG cell lines were determined with MTT assay. Laser scanning confocal mircroscopy and flow cytometry techniques were used to determine the changes of intracellular calcium concentration,CD44V6 protein expression and cell cycle distribution after treatment with tea polyphenols.RESULTS:Three different concentrations dose dependently inhibited the proliferation of PG cell lines. It caused a decline in the proportion of cells in S and G2/M phase, blocked the cell cycle progression in G0/G1 phase and reduced the proliferation index of PG cell lines. Compared with control group, intracellular calcium concentration are gradually increased; the expression of CD44V6 protein are gradually decreased. CONCLUSION:Tea polyphenol could inhibit growth of PG cell lines. The mechanism of anti tumor is associated with the changes of intracellular calcium concentration and the expression of CD44V6 protein.
