摘要
目的 :研究 17β 雌二醇 (E2 )对人宫颈癌HeLa细胞系增殖的影响 ,并探讨其可能的机制。 方法 :应用细胞培养、细胞计数、甲基噻唑基四唑 (MTT)、流式细胞术和激光共聚焦显微镜技术。 结果 :E2 (10 - 6mol L)使细胞生长曲线上移 ;E2 (10 - 8~ 10 - 6mol L)呈剂量依赖性地促进宫颈癌HeLa细胞系的增殖。流式细胞术检测细胞周期发现 ,E2 (10 - 6mol L)可促进细胞周期由G1 期进入S期 ,G1 期的DNA含量由 70 .0 %下降到 5 5 .5 % ,S期的DNA含量由17.3%上升到 2 9.0 %。激光共聚焦检测到E2 (10 - 6mol L)可使细胞内Ca2 +浓度显著升高。 结论 :E2 可以通过调节细胞周期和细胞内Ca2 +浓度水平 。
Objectives:To study the effects of 17β-estradiol on the proliferation of human cervical carcinoma HeLa cells. Methods:Cell culture techniques, growth curves, MTT flow cytometry and laser scanning confocal microscope were used in the study. Results:17β-Estradiol(10 -6 mol/L) makes the curve move upward.17β-Estradiol(10 -8~10 -6 mol/L) significantly stimulated the proliferation of HeLa cells in a dose-dependent manner.17β-Estradiol(10 -6 mol/L) stimulated the proliferation of HeLa cells at G 1/S phases. It blocked G 1 phase from 70.0% to 55.5%, and stimulated S phase from 17.3% to 29.0%.17β-Estradiol (10 -6 mol/L) also increased the release of calcium in HeLa cells. Conclusions:17β-Estradiol can significantly stimulate the proliferation of cervical carcinoma cells possibly by activating cell cycle and stimulating the release of calcium in HeLa cells.
