摘要
目的 研究雌激素 (E2 )对人宫颈癌HeLа细胞系增殖的影响及ERβ在细胞中的表达 ,探讨雌激素影响HeLа细胞增殖的可能机制。方法 采用细胞培养、MTT、流式细胞技术、激光共聚焦显微镜技术、RT PCR技术和DNA测序技术。结果 E2 (10 -8~ 10 -6mol/L)剂量依赖性地促进宫颈癌HeLа细胞的增殖 ,E2 (10 -6mol/L)可促进细胞周期由G1期进入S期 ,G1期的平均DNA含量由 (62 63± 0 42 ) %下降到 (5 1 43± 1 0 1) % ,S期的平均DNA含量由 (2 5 0 3± 1 81) %上升到 (3 2 17± 1 97) %。激光共聚焦检测到E2 (10 -6mol/L)可使细胞内Cа2 +浓度显著升高。RT PCR结果显示HeLа细胞表达ERβmRNA ,大小为 3 88bp ,DNA序列测定证实所得片段为人ERβ的其中一段。 结论 E2 可以通过调节细胞周期和细胞内Cа2 +浓度水平 ,从而调节宫颈癌HeLа细胞的增殖活动 ,并且E2 可能通过与ERβ结合 ,将增殖相关信号传递到细胞内。
Objective To investigate the effects of estrogen on proliferation of human cervical carcinoma HeLa cells and the expression of ERβ to explore the possible mechanisms of cell proliferation Methods The techniques of cell culture,MTT,flow cytometry,laser scanning confocal microscope,RT PCR and DNA sequence assay were used Results Estrogen(10 -8 ~10 -6 mol/l) significantly stimulated the proliferation of HeLa cells in a dose dependent manner Estrogen(10 -6 mol/L) stmulated the proliferation of HeLa cells at G1/S phases Itblocked G1 phase from 62 63±0 42% to 51 43±1 01%,and stimulated S phase from 25 03±1 81% to 32 17±1 97% E2(10 -6 mol/L) also increased the release of calcium in HeLa cells ERβ mRNA was expressed in HeLa cells The result of DNA sequence assay proved that the sequence was some part of human ERβ Conclusions E2 can significantly stimulate the proliferation of HeLa cervical carcinoma cells possibly by activating cell cycle and stimulating the release of calcium And the expression of ERβ protein
