摘要
为探讨mi R-153-3p调控星形胶质细胞升高基因1(AEG-1)对子宫内膜癌细胞上皮间质转化(EMT)的影响,体外培养人子宫内膜癌细胞Ishikawa,分为对照组、mimic NC组(转染mimic NC)、mi R-153-3p mimic组(转染mi R-153-3p mimic)、pc DNA3.1组(转染mi R-153-3p mimic、pc DNA3.1)和pc DNA3.1-AEG-1组(转染mi R-153-3p mimic、pc DNA3.1-AEG-1).结果显示,上调miR-153-3p表达可显著提高Ishikawa细胞凋亡率、细胞中miR-153-3p水平及Caspase-3、E-钙黏蛋白蛋白水平(P <0.05),降低细胞侵袭数、细胞中AEG-1 mRNA及AEG-1、MMP-9、N-钙黏蛋白、波形蛋白、纤连蛋白蛋白水平(P <0.05);在此基础上,上调AEG-1表达可显著逆转上调miR-153-3p表达的影响. miR-153-3p与AEG-1存在靶向关系.综上,miR-153-3p可靶向抑制AEG-1表达,抑制Ishikawa细胞EMT、侵袭行为,并诱导细胞凋亡.
To investigate the effect of miR-153-3p regulating astrocyte elevated gene-1(AEG-1)on the epithelial-mesenchymal transition(EMT)of endometrial cancer cells.Ishikawa human endometrial cancer cells were cultured in vitro and divided into control group,mimic NC group(transfected with mimic NC),miR-153-3p mimic group(transfected with miR-153-3p mimic),pcDNA3.1 group(transfected with mimic and pcDNA3.1)and pcDNA3.1-AEG-1 group(transfected with miR-153-3p mimic and pcDNA3.1-AEG-1).The increase of miR-153-3p expression significantly increased the apoptotic rate of Ishikawa cells,the level of miR-153-3p and the levels of Caspase-3 and E-cadherin proteins(P<0.05),and decreased the number of cell invasion and the levels of AEG-1 mRNA and AEG-1,MMP-9,N-cadherin,Vimentin and Fibronectin proteins(P<0.05).On this basis,the increase of AEG-1 expression can significantly reverse the effect of the increase of miR-153-3p expression.miR-153-3p and AEG-1 had a targeting relationship in Ishikawa cells.In conclusion,miR-153-3p can target and inhibit the expression of AEG-1,inhibit the EMT and invasion behavior of Ishikawa cells,and induce cell apoptosis.
