温阳通络方调控类风湿关节炎成纤维样滑膜细胞上皮间质转化的机制研究

摘要 目的研究温阳通络方对类风湿关节炎(rheumatoid arthritis,RA)成纤维样滑膜细胞(fibroblast-like synovial cells,FLS)上皮间质转化(epithelial-mesenchymal transition,EMT)的调控作用,探讨其作用机制。方法CCK-8实验筛选温阳通络方最佳含药血清浓度;将细胞分为正常关节成纤维样滑膜细胞(human fibroblast-like synovial cells,HFLS)组、人类风湿关节炎滑膜成纤维细胞(human rheumatoid arthritis synovial fibroblasts,MH7A)组、MH7A+温阳通络方含药血清(medicated serum,MS)组,Western Blot检测EMT相关蛋白E-钙黏蛋白(E-cadherin,E-cad)、纤维连接蛋白(fibronectin,FN)和波形蛋白(vimentin,VM)的表达;CCK-8实验检测各组细胞活性;Transwell实验检测各组细胞迁移和侵袭能力。结果96 h时15%温阳通络方含药血清干预后MH7A细胞活性最接近正常细胞的一半,即半抑制浓度(half maximal inhibitory concentration,IC50);MH7A细胞中E-cad的表达水平降低(P<0.05)、FN和VM的表达水平升高(P<0.05),细胞的活性、迁移和侵袭能力增强(P<0.05);温阳通络方含药血清干预MH7A细胞后,细胞中E-cad的表达水平升高(P<0.05)、FN和VM的表达水平降低(P<0.05),细胞的活性、迁移和侵袭能力减弱(P<0.05)。结论温阳通络方可能通过抑制MH7A的EMT现象,抑制滑膜细胞的活性、迁移和侵袭,从而发挥对RA的治疗作用。 Objective To study the regulatory effect of Wenyang Tongluo Prescription(温阳通络方)on the epithelial-mesenchymal transition(EMT)of fibroblast-like synovial(FLS)cells in rheumatoid arthritis(RA)and to explore its mechanism.Methods CCK-8 test was used to screen the optimal serum concentration of Wenyang Tongluo Prescription.The cells were divided into human fibroblast-like synovial cells(HFLS)group,human rheumatoid arthritis synovial fibroblasts(MH7A)group,MH7A+Wenyang Tongluo Prescription medicated serum(MS)group.The expressions of EMT-related proteins E-cadherin(E-cad),Fibronectin(FN)and Vimentin(VM)were detected by Western Blot.Cell activity was detected by CCK-8 assay.The ability of cell migration and invasion was detected by Transwell assay.Results After intervention of 15%serum containing Wenyang Tongluo Prescription at 96 h,the activity of MH7A cells was mostly close to half of that of normal cells,that is,half maximal inhibitory concentration(IC 50).The expression level of E-cad in MH7A cells was decreased(P<0.05),the expression levels of FN and VM were increased(P<0.05),and the cell activity,migration and invasion ability increased(P<0.05).After intervention of MH7A cells with serum containing Wenyang Tongluo Prescription,the expression level of E-cad in MH7A cells was increased(P<0.05),the expression levels of FN and VM decreased(P<0.05),and the activity,migration and invasion ability of cells were decreased(P<0.05).Conclusion Wenyang Tongluo Prescription may inhibit the activity,migration and invasion of synovial cells by inhibiting the EMT phenomenon of MH7A,so as to play a therapeutic role in RA.
