摘要
目的 探讨红丹液通过抑制上皮细胞-间质转化(EMT)防治宫腔粘连的机制,并分析其时效性,为抗纤维化治疗提供新策略。方法 采用100只大鼠建立宫腔粘连模型,分为观察组(红丹液干预)和对照组(无干预)。通过蛋白质印迹(Western blot)技术检测EMT相关蛋白[E-钙黏蛋白(E-cadherin)、波形蛋白(Vimentin)、铁死亡抑制蛋白1(FSP-1)和α-平滑肌肌动蛋白(α-SMA)]的表达,并结合细胞增殖能力(MTT法)评估细胞增殖能力。结果 观察组E-cadherin表达增加,Vimentin、FSP-1和α-SMA表达降低,细胞增殖能力增强,且红丹液干预效果在不同时间点均显著(P<0.05)。结论 红丹液通过调控EMT相关标志物的表达,有效防治宫腔粘连,并显示出明显的时效性,为临床治疗提供依据。
Objective To explore the mechanism by which red lead solution prevents and treats intrauterine adhesions by inhibiting epithelial-mesenchymal transition(EMT),and to analyze its timeliness,providing a new strategy for anti-fibrotic treatment.Methods One hundred rats were used to establish the intrauterine adhesion model and were divided into the observation group(with red lead solution intervention)and the control group(without intervention).The expressions of EMT-related proteins(E-cadherin,Vimentin,FSP-1,α-SMA)were detected by Western blot,and the cell proliferation ability was evaluated in combination with the MTT method.Results In the observation group,the expression of E-cadherin increased,the expressions of Vimentin,FSP-1 andα-SMA decreased,the cell proliferation ability was enhanced,and the intervention effect of red lead solution was significant at different time points(P<0.05).Conclusion Red lead liquid effectively prevents and treats intrauterine adhesions by regulating the expression of EMT-related markers,and shows obvious timeliness,providing a basis for clinical treatment.
