基于多成分HPLC指纹图谱方法的青钱柳质量分析及抗氧化活性研究

Quality and antioxidant activities of Cyclocarya paliurus(Batal.)Iljinsk.:A study based on multi-component high-performance liquid chromatography fingerprint

摘要 目的:通过高效液相色谱(high performance liquid chromatography,HPLC)技术构建青钱柳Cy-clocarya paliurus(Batal.)Iljinsk.的指纹图谱,为其质量评价提供可靠依据,并对其抗氧化活性进行系统研究。方法:采用80%甲醇作为提取溶剂,运用HPLC技术构建青钱柳的指纹图谱,利用《中药指纹图谱相似度评价系统》(2012版)进行相似度分析;采用1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DP-PH)自由基清除法和2,2'-联氮-二(3-乙基苯并噻唑啉-6-磺酸)二铵盐[2,2'-azino-bis(3-ethylbenzothia-zoline-6-sulfonic acid),ABTS]法对15批青钱柳样品的抗氧化活性进行测定。结果:成功构建了15批青钱柳的HPLC指纹图谱,共标定14个共有特征峰,其相似度范围为0.880~0.983。通过与对照品对比,鉴定出8个特征峰成分,包括新绿原酸(峰3)、绿原酸(峰6)、隐绿原酸(峰7)、金丝桃苷(峰8)、异槲皮苷(峰9)、槲皮苷(峰11)、槲皮素(峰13)和山柰酚(峰14)。以槲皮苷(峰11)为参照峰,各共有峰相对峰面积的RSD为24.23%~140.66%,表明不同批次青钱柳样品中部分共有化学成分含量存在显著差异。抗氧化活性测定结果显示,6月份采集的青钱柳表现出更强的自由基清除能力。结论:本研究构建的青钱柳HPLC指纹图谱分析方法操作简便、快速可靠,可作为青钱柳质量控制的科学依据。 Objective:To establish the fingerprint of Cyclocarya paliurus(Batal.)Iljinsk.using high-performance liquid chromatography(HPLC),to provide a reliable basis for its quality assessment,and to systematically investigate its antioxidant activities.Methods:With 80%methanol as the extraction solvent,HPLC was used to establish the fingerprint of Cyclocarya paliurus(Batal.)Iljinsk.,and a similarity analysis was performed using Traditional Chinese Medicine Fingerprint Similarity Evaluation System(version 2012).The 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical scavenging method and the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)radical scavenging method were used to assess the antioxidant activities of 15 batches of Cyclocarya paliurus(Batal.)Iljinsk.samples.Results:The HPLC fingerprints were successfully established for the 15 batches of Cyclocarya paliurus(Batal.)Iljinsk.samples,and a total of 14 common characteristic peaks were identified,with a similarity of 0.880-0.983.Compared with the reference products,eight characteristic peak components were identified,i.e.,neochlorogenic acid(peak 3),chlorogenic acid(peak 6),cryptochlorogenic acid(peak 7),hyperoside(peak 8),isoquercitrin(peak 9),quercitrin(peak 11),quercetin(peak 13),and kaempferol(peak 14).With quercitrin(peak 11)as the reference peak,the relative peak area of each common peak had a relative standard deviation of 24.23%-140.66%,suggesting that there were significant differences in several common chemical components between different batches of Cyclocarya paliurus(Batal.)Iljinsk.samples.Antioxidant activity assay showed that Cyclocarya paliurus(Batal.)Iljinsk.collected in June had a stronger free radical-scavenging capacity.Conclusion:The HPLC method for establishing the fingerprint of Cyclocarya paliurus(Batal.)Iljinsk.in this study is simple,convenient,rapid,and reliable and can be used as a scientific basis for the quality control of Cyclocarya paliurus(Batal.)Iljinsk.
