环境因素对大肥蘑菇菌丝生长的影响 被引量:4

摘要 【目的】研究pH值、温度、光照条件、基质含水量等环境因素对新疆博斯腾湖野生大肥蘑菇组织分离菌株200561-1菌丝的影响,为其人工驯化栽培提供理论依据。【方法】采用组织培养方法,分别研究不同培养基pH值(pH 4.5,5.0,5.5,…,8.5)、培养温度(4,8,12,…,32℃)、光照条件(连续黑暗、12h/12h光暗交替、持续光照)、基质含水量(45%,52%,59%,…,80%)对大肥蘑菇菌株200561-1菌丝生长的影响。【结果】大肥蘑菇菌株200561-1菌丝适宜在中性偏酸环境下生长,pH为6.0~7.0时菌丝表现出良好的生长速度和长势,6.81为其最佳pH值;大肥蘑菇菌丝适宜生长的温度为24~32℃,最佳温度为28.42℃,根据菌丝和培养温度的关系,可将大肥蘑菇菌株200561-1划分为中温偏高型;连续黑暗的培养条件有利于菌丝的生长,这与该菌在自然条件下的生活史相吻合;大肥蘑菇菌丝在基质含水量为66%,73%和80%时均能良好生长,以74.65%为其最佳值,当基质含水量<52%时菌丝不生长,最后萎缩。【结论】新疆博斯腾湖野生大肥蘑菇组织分离菌株200561-1菌丝培养阶段所需的最适pH值为6.81,最适温度为28.42℃,连续黑暗的培养条件有利于菌丝的生长,最适基质含水量为74.65%。 【Objective】This study investigated the effect of environment factors on growth of Agaricus bitorquis strain 200561-1 obtained from wild fruiting body near Bosten Lake by tissue isolation to provide basis for artificial domesticating cultivation.【Method】The effects of pH(4.5,5.0,5.5,…,8.5),temperature(4,8,12,…,32 ℃),light(continuous darkness,12 hlight and 12 hdarkness and continuous illumination),and substrate moisture(45%,52%,59%,…,80%)on mycelial growth were studied using tissue culture technique.【Result】It was suitable for mycelium of Agaricus bitorquis in neutral to acidic environment.The mycelium showed good growth rate and vigor when pH was 6.0-7.0 with the optimum of6.81.The suitable temperature was 24-32℃ with the optimum of 28.42℃.According to the relationship of mycelium and culture temperature,Agaricus bitorquis strains 200561-1 belonged to medium to high temperature type.Continuous darkness conditions was benefit to mycelium growth,which is consistent to the natural life cycle of Agaricus bitorquis.The mycelium got good growth when substrate moistures were66%,73% and 80% with optimal value at 74.65%.The mycelium shrined when the moisture was under52%.【Conclusion】The suitable conditions for mycelium growth of Agaricus bitorquis strain 200561-1 were pH 6.81,temperature 28.42 ℃,continuous darkness,and substrate moisture 74.65%.
