摘要 目的:建立桑椹颗粒中绿原酸的含量测定方法。方法:采用薄层鉴别法(TLC)对制剂中绿原酸进行定性鉴别;以HPLC法测定桑椹颗粒中绿原酸的含量,采用Kromasil C_(18)色谱柱(250 mm×4.6 mm,5μm);使用乙腈-3m L·L^(-1)磷酸水溶液(15∶85)为流动相进行梯度洗脱;流速为0.8 m L·min^(-1);以327 nm为检测波长;柱温为室温。结果:绿原酸的薄层鉴别具有良好的专属性;绿原酸的回归方程:Y=42256X-9219.6,r=0.9994,线性范围为1.300~312.000μg·m L^(-1);平均回收率为96.62%,RSD%值为1.05%。结论:本文建立了桑椹颗粒中绿原酸的HPLC含量测定方法,结果准确,重复性好,可作为桑椹颗粒的质量评价方法。 Objective: To establish a method on content determination of chlorogenic acid component in Mulberry Particles by high performance liquid chromatography (HPLC). Methods: The preparation chlorogenic acid was identified by TLC qualitatively. A high performance liquid chromatography (HPLC) method was used, and Kromasil C,s (250mm×4.6mm,5 μm) column was used, using acetonitrile-3 mL·L-1 phosphoric acid solution (15:85) as mobile phase in a gradient elution mode. Flow rate was 0.8 mL·min-1, detection wavelength was 327 nm, and column temperature was set at indoor temperature. Results: Chlorogenic acid TLC showed good speci- ficity. Chlorogenie acid regression equation was Y=42256X-9219.6 and r=0.9994 in the concentration range of 1.300-312.000 μg·mL-1, and average recovery was 96.62%,and its RSD was 1.05%. Conclusion: The method is accurate, reproducible and suitable to be used for quality assessment of Mulberry Particles.
