Determination of DNJ from Mulberry Leaves by RP-HPLC

摘要 目的:改进反相高效液相色谱-紫外检测法测定桑叶中1-脱氧野尻霉素(DNJ)的定量分析方法。方法:以9-氯甲酸芴甲酯为柱前衍生试剂,采用RP-HPLC法,色谱柱为Agela odyssilC18(250mm×4.6mm,5μm),流动相为V乙腈∶V0.1%冰醋酸=40∶60,流速1.0mL/min,检测波长254nm,柱温30℃。结果:1-脱氧野尻霉素的检测质量浓度在1.158×10-3~2.5mg/mL范围,与峰面积呈良好的线性关系(R2=0.9998),平均回收率为100.9%。结论:本法简便可行,重复性好,准确度高,适合桑叶中DNJ含量的检测。 Objective: To determine DNJ from mulberry leaves by RP-HPLC with precolumn derivatization. Methods: The separation was performed on a Agela odyssil C18 column(250 mmx4.6 mm, 5 μm), with a mobile phase of aeetonitrile-0.1% acetic acid (40:60). The flow rate was 1.0 mL/min and the detection wavelength were set at 254 nm with column temperature at 30℃. Results: The linear range of DNJ were from 1.158×10-3-2.5 mg/mL(R2=0.9998) with recoveries of 100.9%. Conclusion: The method is simple, accurate and responsible for the determination of DNJ from mulberry leaves.
