摘要
目的 建立反相高效液相色谱 (HPLC) 荧光检测测定人全血中丙泊酚浓度的方法。方法 用环己烷提取全血中的丙泊酚 ,以百里酚为内标定量 ,用HPLC 荧光检测器测定丙泊酚含量。结果 本法测定的线性范围为 5~ 30 0 0ng/ml,该方法回收率高 ,重现性好。结论 本方法灵敏 ,选择性强 ,快速、准确地测出全血中微量丙泊酚的含量 。
Objective To develop a method for the determination of propofol concentration in blood by using a reverse phase HPLC and fluorescence detector system,and to measure the blood concentration of propofol during anesthesia.Methods The propofol in blood was extracted with cyclohexane,and thymol was used as the internal standard,determining the concentration of propofol by HPLC fluorescence detector.Results The linear ranges of propofol detected were 5 3000ng/ml.This method was high recovery and fine reproducibility.Conclusion The improved method was a sensitive,rapid,precise and accurate.It is suitable for clinical propofol study. [
