摘要
目的:研究丙泊酚用于神经外科全身麻醉手术患者的药动学。方法:8名神经外科全身麻醉手术患者静脉注射丙泊酚2mg·kg-1后,用高效液相色谱法测定血清中丙泊酚的浓度。用3p97程序拟合计算药动学参数。结果:静脉注射丙泊酚后的药-时曲线符合三室模型;主要药动学参数t1/2α、t1/2β、t1/2γ分别为(0·8±0·41)、(10·2±15·6)、(113·0±47·2)min,Vc、Vd分别为(0·0548±0·0218)、(2·6482±0·6638)L·kg-1,CL为(17·4±4·5)mL·min-1·kg-1,AUC0~∞为(108·92±31·68)mg·min·L-1。结论:丙泊酚的药动学特征与临床麻醉中迅速分布于全身、起效快、作用时间短、苏醒恢复迅速等特点一致。
OBJECTIVE: To study the pharmacokinetics of propofol in neurosurgical patients undergoing general anesthesia.METHODS: 8 neurosurgical patients undergoing general anesthesia were assigned to receive propofol iv(2m ·kg^-1), with their serum concentration of propofol determined by HPLC and pharmacokinetic parameters processed with 3P97 program. RESULTS: The serum concentrationtime curve was in line with three compartment model .The pharmacokinetic parameters were the following: tl/2α, t1/2β and t1/2γ were (0.8± 0.41), (10.2± 15.6) and (113.0± 47.2)min, respectively; Vc and Vd were (0.054 8±0.021 8) and (2.648 2±0.663 8) L · kg^-1, respectively; CL was(17.4±4.5) mL · min^-1·kg^-1 and AUC0-∞ was (108.92± 31.68) mg · min · L^-1.CONCLUSION: Similar to that manifested in clinical anesthesia, the pharmacokinetics of propofol are characterized by rapid distribution throughout body, rapid onset, short duration of action and rapid recovery of consciousness etc.
