摘要 目的观察ATRA对K562细胞的诱导分化作用,探讨c-myc基因表达与诱导分化的关系及可能的作用机制。方法 采用形态学观察、流式细胞术及RT-PCR方法,检测K562细胞向粒系分化特征及c-myc基因的表达。结果 ATRA诱导K562细胞的诱导分化的同时,伴有c-myc基因mRNA水平表达下降。结论ATRA可通过下调c-myc基因表达而诱导K562细胞向粒系成熟方向分化。 Objectives To observe the differentiated characters of K562, to investigate the relationship between c - myc expression and differentiation in K562 cell by all trans - retinoic acid (ATRA) and the possible mechanism. Methods Using morphology (Benzidine dyeing, Wright's dyeing, NSE dyeing and NBT dyeing,) ,flow cytometry and RT - PCR to measure the differentiated characters and c - myc expression in K562. Results After induced by ATRA, K562 cell showed some myelocytic characters and the expression of c - myc mRNA de creased.Conclusion By inhibiting c- myc mRNA expression ATRA can induce K562 to myelocytic differentiation.
