摘要
目的探讨c-myc、hTERT在4-氨基-2-三氟甲基苯基维甲酸酯(ATPR)诱导NB4细胞分化中的作用。方法 NB4细胞按不同处理方式分为以下各组:以RPMI 1640培养基作空白对照,乙醇作溶剂对照,全反式维甲酸(ATRA,1μmol/L)作阳性对照,ATPR(1μmol/L)处理组。采用RT-PCR法和Western blot法检测4d后c-myc与hTERT的mRNA及蛋白的表达变化,并选取ATPR组处理0、1、2、3、4、5 d时的细胞进行时效性研究。结果 ATPR作用NB4细胞4d后c-myc与hTERT的mRNA及蛋白的表达显著下降。时效性研究发现随着时间的延长,c-myc与hTERT的mRNA及蛋白的表达亦呈下降趋势。结论通过抑制c-myc进而抑制hTERT及端粒酶活性可能是ATPR诱导NB4细胞分化的机制之一。
Objective To explore the effects of c-myc,hTERT on differentiation of NB4 cells induced by 4-amino-2-trifluoromethyl-phenyl retinate(ATPR).Methods The mRNA and protein expressions of c-myc,hTERT were detected by RT-PCR and Western blot with differentiation of NB4 cells induced by ATPR for 4 days and further to conduct the analysis of the time-effects.Results The mRNA and protein expressions of c-myc,hTERT were significantly decreased after treatment with ATPR on NB4 cells for 4 days,and the change of expression had time-dependent(P0.01).Conclusion ATPR probably inhibits the activation of c-myc and hTERT to induce differentiation of NB4 cells.
